Pediatric Immune Response and Improving Vaccines

Over 7 million infants die each year due to infections, many of which are vaccine-preventable. The causes
of reduced immunity in infants are ill defined. The immunosuppressive in utero environment needed for
maternal-fetal tolerance may influence subsequent immune responses in newborns. We have identified
myeloid-derived suppressor cells (MDSC) as a plausible mechanism for reduced immunity in infants.
MDSC frequencies are elevated in cord blood, decline in infants with age and are potent inhibitors of
T cell proliferation. Understanding the role MDSC play in infant immunity could improve vaccine responsiveness
in newborns and reduce mortality due to early-life infections.
